Objective To isolate and culture and purify epidermal cells from rat cervical-loop, and to observe the expression of amelogenin in epidermal cells. Methods Tissues obtained from lower incisor of postnatal 2-day Wistar rats′cervical-loop were cultured in the Dulbecco minimum essential medium(DMEM)-F12 medium. The cervical-loop epidermal cells grew confluent after purified from fibroblast-like cells by differential digestion. Then the purified cells were immunocytochemical stained with antiamelogenin antibody. Results The primary culture was exhibited positive staining to amelogenin while the control group was negative. Conclusion The epidermal cells isolated from rat cervical-loop epithelium have the potentiality to differentiate into ameloblasts.
