目的探究灵芝醇提取物在体内外的抗肿瘤活性。方法用灵芝醇提取物处理6种人肿瘤细胞系(表皮癌A431、胃癌BGC823、直肠癌Caco2、卵巢癌ES-2、肺癌NCI-H460、结肠癌SW620),观察细胞的生长形态、绘制生长曲线、进行MTT实验及平板集落形成实验。分别ig给予胃癌BGC823裸鼠50、100、150 mg·kg~(-1)灵芝乙醇提取物,计算瘤体积、抑瘤率及脏器系数。结果体外实验中,灵芝提取物对表皮癌A431、胃癌BGC823、直肠癌Caco2、卵巢癌ES-2、肺癌NCI-H460、结肠癌SW620细胞的半数抑制浓度分别85.86、193.43、51.32、168.72、82.61、212.98μg·m L~(-1)。体内实验中,灵芝提取物50、100、150 mg·kg~(-1)的抑瘤率分别为32.6%、43.5%、47.2%。结论灵芝提取物抑制表皮癌A431、直肠癌Caco2、肺癌NCI-H460细胞的增殖作用较强,且浓度大于40μg·mL~(-1)时,抑制增殖的作用较明显,体外还能抑制胃癌BGC823的生长。
