高温对人舌鳞癌细胞中nm23-H_1基因表达影响的实验研究

目的 研究高温治疗癌症与转移抑制基因nm23- H1 之间的关系,探讨高温抑制肿瘤细胞侵袭及转移的机理。方法 对人舌鳞癌Tca8113 细胞行体外43 ℃加温,采用电镜、免疫组织化学及流式细胞术等方法,分析加热后舌癌细胞超微结构的变化及nm23- H1 表达量的改变。结果 加温使Tca8113 细胞中细胞器增多,形态趋向良性分化。在加热43 ℃时随加热时间延长,细胞中nm23- H1 表达量增加,在加热30 min 时达到最高,此变化不随加热后培养时间的增加而改变。结论 高温能使肿瘤细胞中nm23- H1 表达量增加,并使细胞形态出现一定分化,从而抑制肿瘤细胞的侵袭、转移能力。43 ℃加热30 min 可能是临床治疗舌鳞癌的理想位点。
英文摘要
Objective To investigate the mechanism of hyperthermia treatment of tongue cancer by observing the relation of expression of nm23- H1 gene and the infiltration and metastasis of the cancer. Methods Tongue squamous carcinoma cells(Tca8113) heated in vitro was observed and analysed through transmission electron microscope( TEM), immunohistochemistry, flow cytometry, then determined the quantities of nm23- H1 and ultrastructure. Results The organelles increased after heated by hyperthermia. In the heated cells, the expression quantities of nm23- H1 increased, but this effect couldn′t be changed as the cultured time prolonged. Conclusion Hyperthermia can suppress cancer cell invasion and metastasis by adjusting the expression of nm23- H1. It is the optimal time for hyperthermia treatment to heat 30 minutes at 43 ℃.
