急性髓细胞白血病(AML)是一种常见血液系统恶性肿瘤,但AML患者的总体化疗效果差、长期生存率低,提高其化疗效果及改善预后成为目前临床亟待解决的难题之一。目前多项研究表明,AML患者的化疗效果差和预后不良可能与AML耐药关系密切。AML耐药不仅与AML细胞自身的生物学特征相关,而且骨髓微环境在AML耐药中亦发挥重要的作用。骨髓微环境不但可以通过分泌多种细胞因子,以及细胞与细胞的接触,保护AML细胞免受化疗药物杀伤,而且可以通过支持白血病干细胞(LSC)存活,调节相关信号通路以介导AML细胞耐药。深入探讨骨髓微环境介导AML细胞的耐药机制,可为靶向干预AML细胞耐药,提高AML化疗疗效奠定基础。

英文摘要

Acute myeloid leukemia (AML) is a common hematopoietic malignant tumor. AML patients have poor chemotherapy effect and low long-term survival rate. To improve the chemotherapy effect and prognosis of AML patients is currently a clinical problem to be solved. The reason of poor chemotherapy effect and prognosis may be closely related to AML drug resistance. AML resistance is not only related to the biological characteristics of AML cells, but also due to bone marrow microenvironment. Bone marrow microenvironment plays an important role in AML resistance. To explore the mechanisms of bone marrow microenvironment mediating drug resistance, lays a foundation for targeted intervention to improve the therapeutic effect of AML chemotherapy.
