To establish and optimization the extraction process of polysaccharide from Polygonatum leaves, the polysaccharide was extracted by the water extraction method, and the content of polysaccharide was determined by the anthrone sulfuric acid method. Based on the yield rate of polysaccharide and the extraction ratio, the three-factor and three-level Box-Behnken response surface methodology(RSM) was used to optimize the extraction process of polysaccharide from Polygonatum leaves by single factor test. The best extraction process of polysaccharide from Polygonatum leaves was: extraction temperature 75 ℃, the ratio of liquid-material 15∶1, extraction time 2.5 h. Under this condition, the extraction ratio was (47.65±0.16) %, and the polysaccharide yield rate was (7.21±0.073) %. The water extraction process of polysaccharide from polygonatum leaves was stable and feasible, and also has a high yield rate.
