In this paper, four kinds of soil total DNA extracting methods are applied to soil sample from Xichang, Sichuan, an available region for Jatropha curcas L. growth and through comparison of extracting effects, we find improved methods, method 1 and method 4, show better extracting results than method 2 and method 3. Moreover, the results of multiplex-PCR amplification show the soil total DNA extracted by method 1 and method 4 can obtain higher ratio of fungi DNA, suggesting the DNA extracted by this two methods is more representative. However, only method 4 can successfully extract total DNA of the soil samples from Hainan Province and Yunnan Province, the low DNA purity and yield meaning further modification is needed.
