根据基因芯片的数据筛选出Osa-miR446及靶基因作为研究对象。生物信息学分析表明,Osa-miR446有4个候选靶基因,通过RACE技术对这些候选靶基因进行鉴定,发现只有LOC_Os03g56930.1(APP1基因)可得到理想的剪切条带,测序后比对可知该剪切位点位于APP1基因的内含子区域(第1036-1037个碱基)。随后,通过克隆获得APP1基因,信息学分析可知该基因开放阅读框长度为711bp,是一个编码237个氨基酸的新的未定性的疏水性蛋白质。Southern杂交结果显示APP1在水稻基因组中为单拷贝基因。 利用荧光定量PCR分析了Osa-miR446及其靶基因APP1在非生物胁迫中的表达模式,结果表明Osa-miR446与APP1基因的表达均呈负相关,说明Osa-miR446确实可通过剪切APP1基因的转录产物来对非生物胁迫应答。

英文摘要

We researched Osa-miR446 and its target gene APP1 on the basis of gene microarray. According to bioinformatics analysis, Osa-miR446 might have four target genes, but only LOC_Os03g56930.1 (named APP1) could be identified as one of target genes of Osa-miR446 via the RACE experiment. APP1 had an open reading frame of 711bp,encoding an undefined hydrophobicity protein with 237 amino acids. The Southern experiment showed that APP1 was a single-copy gene in rice genome. Expression profile under stressed of Osa-miR446 and APP1 was analyzed. Overall, the results indicated miR446 could regulate APP1 via splicing the transcriptions under stresses and they both played roles in stress response.
