猪SOCS7基因克隆、组织表达图谱分析及剪接变体鉴定

以家猪肌肉来源cDNA为模板, 克隆了SOCS7的编码区cDNA全长并发现了三种剪接变体(命名为: SOCS7 v1、SOCS7 v2和SOCS7 v3). 家猪SOCS7编码区cDNA全长为1785bp, 由9个外显子组成, 编码含581个氨基酸残基的蛋白质. 家猪SOCS7基因的三种剪接变体分别编码含547、520、512个氨基酸残基的蛋白质. 通过RT PCR方法, 本研究对家猪SOCS7基因及其剪接变体的组织表达图谱进行了分析, 结果显示: SOCS7在精巢、小脑、背肌、腹肌、股肌、大脑、下丘脑、心脏、肝脏、脾脏、小肠等组织中均有表达, 在精巢中表达信号最强. SOCS7 v1和SOCS7 v2除大脑外的19个检测组织中均能检测到较强的表达信号, SOCS7 v3在除精巢外的19个组织中均有表达. 以上实验结果为探究家猪SOCS7基因的生理功能奠定了基础.

The full length cDNA of SOCS7 was cloned using pig muscle cDNA as a template, and identified three novel transcript variants (named SOCS7 v1, SOCS7 v2 and SOCS7 v3). The full length cDNA of SOCS7 encodes a protein of 581 amino acids, while the three novel transcript variants (SOCS7 v1, SOCS7 v2 and SOCS7 v3) encode proteins of 547, 521, 512 amino acids respectively. Using RT PCR method, we further analyzed the tissue distribution of SOCS7 and its transcript variants. The results showed that: the full length SOCS7 protein is expressed in the testis, cerebellum, back muscles, abdominal muscles, femoral muscle, brain, hypothalamus, heart, liver, spleen and small intestine. While the SOCS7 v1 and SOCS7 v2 are expressed in 19 tissues except cerebrum; SOCS7 v3 is also expressed in 19 tissue examined except testis. These results lay the molecular basis for further exploration of physiological roles of SOCS7 in pigs.
