双孢蘑菇原生质体融合子的特性与分子鉴定

采用原生质体技术, 以双孢蘑菇主推品种As2796和浙农1号为亲本, 获得性状优异的融合子Z 3和Z 7, 二者均可在34 ℃生长良好, 菌丝纤维素酶活性和漆酶活力均比As2796高,酯酶同工酶谱显示与亲本差异明显, ITS序列证实二者与As2796同源性分别为997%, 993%.栽培试验得出新菌株Z 3和Z 7的产量分别比As2796高125% 和51%.从其经济性状的综合指标看, 可考虑进一步的区域试验与示范推广栽培.
英文摘要
Two fusants, named Z 3 and Z 7, were screened out from protoplast fusion between two main cultivated strains Agaricus bisporus 2796 and Zhe Nong No.1. Results showed both of them can grow at 34 ℃, the activities of cellulose enzyme and laccase were higher than As2796. Besides, it demonstrated big differences by analyzing EST isoenzyme banding patterns. Further ITS sequences homologies were identified with 997% and 993%, respectively. Cultivated experiment proves that the strains of Z 3 and Z 7 increased the yields with 125% and 51%, respectively. It can be promoted after further field experiments, regarding its good economic indicator.
