本文以RCAR1为诱饵蛋白通过酵母双杂交(Y2H)技术筛选出了DWD基因编码的蛋白,同时在用ABI2参与筛选的过程中,同样得到了DWD这个基因. 进行了pull down 实验验证它们之间的相互作用,实验结果显示:在体外,RCAR1、RCAR3与DWD之间确实存在相互作用,同时ABI2与DWD也存在相互作用. 其后对DWD结构的进行分析和实验,发现ABI2和DWD的相互作用区域集中在AtDWDC端,大概是145~350位氨基酸之间的WD40结构域里.
英文摘要
AtDWD(WD40) protein was obtained by using yeast two hybrid screening with the bait of RCAR1 in former work. Further studies demonstrated that ABI2(a kind of PP2C) could interact with AtDWD. Due to the deficiency of the yeast two hybrid technology, this study used GST pull down to determine the interaction between ABI2 and AtDWD. The results showed that the interaction between AtDWD and ABI2 was exist. In addition the study showed that 145~350 aa in the C terminus of AtDWD interacted with ABI2 (PP2C).
