The Fusarium solani R13, isolated from Rheum palmatum L. lived in plateau, was studied in this research. In order to raise the emodin production of R13 and explore the synthetic route of emodin, the ways of regulating the initial pH of the fermentation broth and adding precursors of the anthraquinone synthetic pathway were adopted, and the quantitative analysis was made by using of HPLC. The result showed that the production of emodin was promoted when adding propionic acid and malonic acid, and the effects were concentration dependent. It could be preliminary inferred that the synthetic pathway of emodin produced by F . solani R13 belonged to anthraquinones biosynthesis route. And the production of emodin achieved 3908 mg/L, which lay the foundation for practical application of emodin produced by F . solani R13.
