酸性染料比色法测定百部流浸膏中的总生物碱

目的采用酸性染料比色法测定百部流浸膏中总生物碱的含量,为百部流浸膏质量标准的提升提供依据。方法取0.1 mL供试品溶液,置具塞离心管中,加入5 mL溴甲酚绿、7.0 mL pH4.6的邻苯二甲酸氢钾缓冲液、6 mL二氯甲烷溶液,密塞,涡旋2 min,静置30 min,取二氯甲烷层溶液置10 mL量瓶中,用二氯甲烷定容,在425 nm处测定吸光度。结果对叶百部碱的标准曲线为:Y=949.76X-10.756(r~2=0.9975),线性范围为53.125~850μg·mL~(-1);平均回收率为105.20%,RSD=4.16%。13份百部流浸膏样品的总生物碱含量为14.01~74.56 mg·mL~(-1),建议百部流浸膏中总生物碱的含量不低于11.21 mg·mL~(-1)。结论酸性染料比色法具有良好的专属性、精密度和稳定性,操作简便,可作为百部流浸膏生物碱含量测定的有效方法。
