目的采用HPLC法测定人尿液中莫吉司坦活性代谢物M1的含量。方法采用Inertsil C8-3色谱柱(250 mm×4.6mm,5μm),流动相为乙腈-0.01 mol·L-1KH2PO4(pH6.8)(30∶70),流速为1 mL·min-1,萘普生为内标,检测波长为230nm,柱温为室温。结果 M1的线性范围为2.5~1×103μg·mL-1,最低定量限为2.5μg·mL-1。批内精密度和批间精密度均小于4%,方法回收率为95.35%~102.2%,萃取回收率为99.1%~99.7%。结论所用方法具有快速简便、灵敏准确的特点,可满足M1临床药物浓度测定的需要。
