目的 探讨不同浓度17β-雌二醇(17β-E2)对二氧化硅(SiO2)刺激的小鼠肺成纤维细胞表达Caveolin-1和Ⅲ型胶原的影响。方法 将体外培养的小鼠肺成纤维细胞分5组:空白对照组,SiO2(100 mg/L)组,SiO2(100 mg/L)+不同剂量17β-E2组(10-8、10-7、10-6 mol/L),17β-E2作用48 h后收集各组细胞,应用免疫组化法检测Caveolin-1和Ⅲ型胶原表达。结果 SiO2 处理组小鼠肺成纤维细胞与空白对照组比较Caveolin-1表达降低(P2+ 17β-E2处理组随着17β-E2剂量增大Caveolin-1表达明显升高(PP2组与空白对照组比较增高(P2处理组均低于SiO2组(P2与Caveolin-1的表达呈正相关(r=0.926,Pr=-0.914,Pr=-0.887,P2可能通过上调肺成纤维细胞Caveolin-1 的表达,抑制Ⅲ型胶原表达而发挥抗纤维作用。

Objective To observe the effects of different concentration of 17β-estradiol (17β-E2) on the expression of Caveolin-1 and type Ⅲ collagen in the mouse lung fibroblast stimulated by SiO2. Methods Fibroblast cells were devided into five groups: blank control group, SiO2 (100 mg/L) group and SiO2 (100 mg/L)+ different concentration of 17β-E2 (10-8,10-7,10-6 mol/L) groups. After treated with different concentration of 17β-E2, for 48 h, the cells were collected, then the expression of Caveolin-1 and of type Ⅲ collagen were examined with immunohistochemistry method. Results Compared with the blank control group, the expression of Caveolin-1 in mouse lung fibroblasts treated by SiO2 significantly decreased (P 2+17β-E2 group significantly increased as 17β -E2 dose increased (P P 2 group were evidently higher than that in the blank control group, while which in 17β-E2 groups decreased significantly, when compared with SiO2 group (P 2 increased the expression of Caveolin-1 and decreased the expression of type Ⅲ collagen in the dose dependent manner. Correlation analysis showed that 17β-E2 was positive correlated with the expression of Caveolin-1 (r=0.926, P r=-0.914, P r=-0.887, P 2 may inhibit the expression of type Ⅲ collagen by up-regulating the expression of Caveolin-1 in mouse lung fibroblast cell to play a role in the resistance of the lung fibrosis.
