中文摘要

为了理解细胞信号转导网络作为非线性动态自治系统的复杂性、系统内在的控制机理和系统的鲁棒性,提出从信息科学和控制工程的观点出发研究细胞网络的系统动态特性。以NF-κB信号转导网络为例,利用动态敏感性分析和非线性动力学分析等方法,研究细胞信号转导网络的参数敏感性和系统输出的振荡特性。仿真结果表明,8个关键的反应速率常数是系统输出信号NF-κBn的敏感参数,IKK的负反馈调节作用显著影响着系统输出NF-κBn的振荡行为。

英文摘要

In order to understand the complexity of cellular signaling pathways as nonlinear autonomous systems, their regulation mechanisms and robustness properties, the system dynamic characteristics of cellular networks were analyzed using perspectives of engineering and information sciences. Taking the NF-κB signaling transduction networks as an example, dynamic sensitivity analysis, nonlinear dynamic analysis were explored to study the parametric sensitivities and oscillation patterns with respect to the system output of cell signal transduction networks. The simulation results demonstrated that the output signal NF-κBn is relatively sensitive to eight key rate constants and the oscillation behavior of NF-κBn is obviously affected by the negative feedback function of IKK.
