在生理pH下,采用光谱法、电化学、化学热力学和粘度法等方法,利用中性红作分子探针,研究了刚果红与鲱鱼精DNA之间的作用方式,获得了结合比、结合常数和热力学函数值。结果表明,刚果红与DNA之间以嵌插方式发生作用。刚果红与DNA之间的结合比为nCR:nDNA=1:1,结合常数为KӨ25℃=4.50×103 L/mol。25℃时,ΔrHmӨ =7.04×104 J/mol,ΔrSmӨ=305.88 J/(mol•K), ΔrGmӨ =-2.08×104 J/mol。刚果红与DNA之间的作用为熵驱动。

英文摘要

In this paper, all of the studies were carried out under the condition of physiological pH. The interaction between Congo red and DNA was studied by spectroscopic, electrochemical, thermodynamic and viscometry methods, using Nature red as molecular probe. Binding stoichoimetry, binding constant and the corresponding thermodynamic parameters were obtained. The results indicated that the interaction of Congo red with DNA is intercalation binding. The binding stoichoimetry of nCR:nDNA=1:1, and the binding constant is KӨ25℃=4.50×103 L/mol. The thermodynamic parameters at 25℃are ΔrHmӨ =7.04×104 J/mol, ΔrSmӨ=305.88 J/(mol•K), ΔrGmӨ =-2.08×104 J/mol respectively. Entropy is driven force.
