中文摘要

脱落酸(ABA)受体在调控植物非生物抗逆过程中起重要作用,因此,研究其相互作用蛋白质对认识受体的调控网络具有重要意义。本研究采用酵母双杂交(Y2H)、Pull down及BiFC实验方法研究蛋白质激酶CARK4与ABA受体RCAR12之间的相互作用。酵母双杂交(Y2H)试验显示,缺失氮端的CARK4(CARK4ΔN)与RCAR12具有明显的相互作用。体外Pull down试验进一步得出CARK4可以直接与RCAR12相互作用。为了确定CARK4与RCAR12在植物体内是否也存在相互作用,利用农杆菌介导的烟草叶片瞬时表达来进行BiFC实验,结果表明在烟草中CARK4也可以直接与RCAR12相互作用。研究结果表明,CARK4与RCAR12在植物体内、体外均具有相互作用。

英文摘要

Abscisic acid receptors play a pivotal role in regulation of environmental stress, its network is fundamental to elicit ABA pathway. In this research, Yeast two- hybrid system (Y2H), Pull down and Bimolecular fluorescence complementation (BiFC) assays were used to study the interaction between CARK4 and RCAR12. The result from Y2H showed an interaction of CARK4ΔN with RCAR12. The same from Pull down were confirmed in vitro. BiFC result indicated that CARK4 interacted with RCAR12 in Nicotiana benthamiana. Taken together, we can conclude that the interact on of CARK4 with RCAR12 is confirmed in vitro and vivo.
