Herpes virus can cause many diseases, such as keratitis, cold sores, herpes zoster, genital herpes, and encephalitis, etc. Aciclovir and related nucleoside analogues are gold standard molecules for treatment of herpetic diseases in clinical over the past decades. Here, the recent progress of anti-herpes virus nucleoside agents are summarized in this review, including structure characteristic, antiviral mechanism and clinical application.
