目的观察蜂王浆(RJ)对H_2O_2诱导的小鼠胚胎成纤维细胞(NIH-3T3)损伤的修复作用,并探讨其可能的分子机制。方法以20μg·mL~(-1) H_2O_2处理NIH-3T3细胞建立细胞损伤模型,用不同浓度的蜂王浆处理模型细胞,检测各组细胞的增殖活性、β-半乳糖苷酶(SA-β-Gal)染色阳性细胞率、细胞凋亡情况以及细胞衰老相关基因的变化。结果 NIH-3T3细胞经H_2O_2处理后增殖能力下降,SA-β-Gal阳性率升高,p16、p21、p53mRNA过高表达;RJ处理后上述指标均有所改善。结论 RJ能延缓H_2O_2所致的细胞损伤和衰老。其机制可能和RJ在翻译前的环节抑制p16、p21、p53基因的表达并通过p16-cyclinD/CDK-RB和p53/P21信号通路调控细胞周期及凋亡程序有关。
