沙棘甾醇对酒精性胃黏膜损伤的保护作用

目的探讨沙棘甾醇对急性酒精性胃黏膜损伤的保护作用。方法将112只SD大鼠,随机均分为对照组、模型组、橄榄油组及沙棘甾醇低、中、高剂量组(100、200、400 mg·kg~(-1))和盐酸雷尼替丁组(50 mg·kg~(-1))。对照组及模型组的大鼠灌胃0.9%生理盐水,橄榄油组的灌胃橄榄油,盐酸雷尼替丁组的灌胃盐酸雷尼替丁,其余各药物组的按剂量溶于橄榄油后灌胃,连续给药7 d,每日1次。在末次给药2 h后,除空白对照组外,其余各组按1 mL·(100 g)~(-1)灌胃75%乙醇,构建急性胃黏膜损伤模型,采集样本,并在透射电镜下观察病理切片。检测大鼠血清中超氧歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)、谷胱甘肽过氧化物酶(GSH-Px)的含量。采用酶联免疫吸附试验法测定血清中肿瘤坏死因子-α(TNF-α)和胃动素(MTL)的含量。结果与模型组比较,沙棘甾醇中、高剂量能显著提高大鼠血清中SOD的活力,降低MDA的含量,沙棘甾醇中、高剂量能有效提升GSH-Px的活力,但无统计学意义;沙棘甾醇各剂量组能显著降低大鼠血清中TNF-α的含量,提高MTL的含量。结论沙棘甾醇能够不同程度地降低乙醇诱导的胃黏膜损伤,具有一定的胃黏膜保护作用。
