目的研究白藜芦醇(RSV)诱导DT40细胞DNA损伤的作用机制。方法采用MTT试验检测RSV对细胞增殖的影响;利用染色体畸变试验分析RSV对细胞的DNA损伤作用以及抗氧剂N-乙酰半胱氨酸(NAC)对RSV诱导DNA损伤的干扰作用。结果 50μmol·L~(-1)RSV连续处理16 h后可诱导DT40细胞的DNA损伤;RSV对DT40细胞的增殖呈剂量依赖性抑制作用,1 mmol·L~(-1)NAC预处理1 h后可提高DT40细胞的生存率,同时也可抑制RSV诱导的DNA损伤。结论 RSV可诱导DT40细胞的DNA损伤,其作用机制可能与RSV的氧化损伤相关。
