阿托伐他汀对大鼠脑出血后脑组织中HMGB1与神经血管再生的影响

目的观察阿托伐他汀对大鼠脑出血后714 d脑组织中高迁移率族蛋白1(HMGB1)、血管内皮生长因子(VEGF)、神经生长因子(NGF)及新生神经血管的影响,探讨阿托伐他汀保护脑出血后脑神经的作用机制和途径。方法 45只健康雄性大鼠随机均分为阿托伐他汀组、脑出血组、假手术组(第7、10、14天3个时间点,每个时间点5只)。阿托伐他汀组于术后予以阿托伐他汀溶液80 mg·kg~(-1),每天1次;假手术组、脑出血组ig等剂量生理盐水。分别检测第7、10、14天时神经功能缺损评分,用蛋白印迹法检测HMGB1、VEGF、NGF的表达情况,免疫组织化学检测第14天时神经血管的新生情况。结果大鼠脑出血24 h后出现明显偏瘫症状,脑出血组大鼠的神经功能缺损评分较假手术组高;阿托伐他汀组大鼠神经功能缺损评分较脑出血组显著降低,提示阿托伐他汀可促进神经功能的恢复;与脑出血组比较,第14天时阿托伐他汀组脑组织中HMGB1、VEGF、NGF蛋白的表达增多,血肿周围新分化的神经元的细胞数较出血组增多,同时周围新生血管明显增多。结论脑出血后,给予阿托伐他汀可上调HMGB1、VEGF、NGF,促进神经和血管的再生,起到了脑保护作用。
