白藜芦醇对脂多糖诱导细胞炎性衰老的抑制作用及机制

目的研究白藜芦醇对脂多糖(LPS)诱导的细胞炎性衰老的抑制作用,并探讨其作用机制。方法培养HEK293细胞,加入LPS诱导细胞炎性衰老,再加入白藜芦醇干预。取培养后的细胞上清液,分别检测半胱天冬酶1(Caspase-1)、肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)的含量;采用实时荧光定量PCR及Western Blot法分别检测SIRT1 mRNA及NLRP3蛋白的表达;采用SA-β-gal染色检测细胞的衰老情况。结果与LPS组比较,白藜芦醇组细胞中Caspase-1、TNF-α及IL-1β的含量减少,NLRP3蛋白的表达下降,细胞焦亡受到明显抑制; S-A-β-gal检测表明细胞衰老得到一定程度缓解; SIRT1的表达水平显著上升。结论白藜芦醇可以减缓细胞炎性衰老;这一抗衰老作用可能与上调SIRT1的表达进而抑制细胞焦亡有关。
