紫芸解毒止痛酊中红花的质量控制及其促骨髓间充质干细胞增殖的作用

目的对紫芸解毒止痛酊(ZYD)中的关键药材红花进行DNA条形码和HPLC化学质量控制,并探究羟基红花黄色素A(HSYA)在促骨髓间充质干细胞(BMSCs)细胞增殖中的作用。方法采用ITS2 DNA条形码技术鉴定中药材红花并分析其与近缘物种的序列差异; HPLC法测定5个批次的ZYD中HSYA的含量; CCK-8法测定BMSCs的细胞相对增殖率。结果 ZYD中红花的ITS2序列与NCBI数据库结果一致,系统进化树构建后发现其与近缘物种各自聚类成分支; 5个批次的HSYA含量分别为23. 72±0. 44、24. 49±0. 15、25. 23±0. 51、23. 52±0. 21、23. 44±0. 42μg·m L~(-1)(n=6),RSD分别为1. 87%、0. 65%、2. 06%、0. 89%、1. 82%;与对照组比较,CCK-8法检测到ZYD、HSYA、红花提取液(HH)组分处理BMSCs 48 h后细胞的相对增殖率均大于100%,有显著促进细胞增殖的作用。结论 ITS2 DNA条形码和HSYA可作为ZYD中关键药材红花质量控制的重要指标,ZYD、HH的促BMSCs细胞增殖作用与HSYA的成分功效有关。
