新型近红外吲哚七甲川菁染料的合成与肿瘤靶向性研究

目的合成两个不同N-取代基的近红外吲哚七甲川菁染料(ICP-15、ICP-16),研究其光声成像能力和肿瘤靶向性。方法采用间羟基苯甲酸与甲醇成酯,再与1,2-二溴乙烷缩合得3-(2-溴乙氧基)苯甲酸甲酯,水解可得3-(2-溴乙氧基)苯甲酸,将酯和酸分别与2,3,3-三甲基-3H吲哚反应制得季铵盐,进而与2-氯-3-羟次甲基环己烯醛缩合制得目标化合物ICP-15、ICP-16。测定目标化合物的最大吸收波长、发射波长。通过建立体内、外模型,以吲哚菁绿(ICG)为对照,对比研究ICP-15、ICP-16的光声成像能力和肿瘤靶向性。结果制备并确证了ICP-15、ICP-16的化学结构,均未见文献报道,其最大吸收和发射波长均在近红外区;体外研究显示:在相同实验条件下,ICP-15、ICP-16的光声信号强度均高于ICG,且其光声信号强度与浓度具有良好的相关性;体内研究显示:ICP-15、ICP-16具有一定的肿瘤靶向性。结论新型吲哚七甲川菁染料ICP-15、ICP-16有望成为光声成像造影剂且具有一定的肿瘤靶向性。
