目的研究槲皮素对糖尿病合并脑缺血再灌注损伤大鼠的神经保护作用及其机制。方法采用高脂饮食联合低剂量链脲佐菌素建立SD大鼠2型糖尿病模型,随机分为假手术组、模型组、尼莫地平组、胰岛素组和槲皮素组;采用线栓法制备大脑中动脉阻塞再灌注模型,再灌注的同时ip给药,每日1次(胰岛素每日2次),假手术组和模型组给予等体积的生理盐水,连续3 d。3 d后,检测血糖,进行神经功能评分,TTC染色测定脑梗死体积,HE染色观察脑组织形态,ELISA检测脑皮质超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量。结果模型组大鼠脑皮质SOD的活性显著降低,MDA的含量显著升高,表明造模成功。治疗3 d后,槲皮素可降低糖尿病大鼠的血糖,减少大鼠的脑梗死体积,减轻神经功能缺损,降低神经功能评分,增强存活神经元的密度;同时可以提高脑皮质中SOD的活性,降低脑皮质MDA的含量。结论槲皮素对糖尿病合并脑缺血再灌注损伤大鼠具有神经保护作用,其作用可能跟降低血糖及调节抗氧化途径有关。
