目的研究α-细辛脑亚微乳对阿尔茨海默症(AD)大鼠空间学习记忆能力的改善作用。方法给予SD大鼠三氯化铝(20 mg·kg~(-1),ig)联合D-半乳糖(180 mg·kg~(-1),ip)建立AD模型,将造模成功的大鼠随机分为细辛脑亚微乳低、高剂量(10、25 mg·kg~(-1))组、盐酸多奈哌齐(1. 75 mg·kg~(-1))组及模型组,每日给药1次,连续28 d;采用Morris水迷宫空间探索法测定大鼠给药前、后穿越平台的次数;采用HE染色法观察大鼠海马区的组织形态;采用免疫组化染色法检测β-淀粉样蛋白(Aβ1-42)、Tau蛋白的表达水平;测定大鼠脑内乙酰胆碱(ACh)、乙酰胆碱酯酶、胆碱乙酰转移酶的含量。结果与模型组比较,α-细辛脑亚微乳高剂量可显著增加AD大鼠穿越平台的次数,其作用可能与减少大鼠海马区神经元的缺失,降低Aβ、Tau蛋白的表达水平,增加脑组织中ACh的含量有关。结论α-细辛脑亚微乳可显著改善AD大鼠的认知缺陷与记忆障碍,具有治疗AD的潜在应用前景。
