壳聚糖季铵盐对镁铝氢氧化物所载药物稳定作用的研究

目的制备具有理想包封率的载药镁铝氢氧化物纳米复合物,并进行体外表征。方法通过测定纳米复合物的粒径、PDI、Zeta电位、包封率、载药量来筛选处方;考察按最佳处方制备的纳米复合物的微观形貌、体外释放行为及细胞毒性。结果按最佳处方所得纳米粒的粒径为371.1 nm,Zeta电位+30.8 mV,包封率64.35%,载药量16.26%;包封率明显改善。该纳米复合物含有0.38%硫元素,证明载体表面吸附了硫酸葡聚糖;纳米复合物的形态为棒状或椭球形。药物在酸性介质中较中性介质中释放更快,充分体现了镁铝氢氧化物的pH依赖性溶解特性,可应用于pH刺激响应性智能给药系统。游离药物和制剂对RAW 264.7细胞的毒性均呈浓度相关性,且甲氨蝶呤等效浓度为0.05~5μg·mL~(-1);制剂组的毒性小于游离药物组。结论成功制备了具有较高包封率的表面改性载药镁铝氢氧化物纳米复合物。
