参归仁合剂对产后抑郁大鼠雌激素和额叶p-CREB及p-ERK1/2的影响

目的探讨补虚化淤方药参归仁合剂对产后抑郁大鼠模型雌激素(E_2)的水平、额叶环磷腺苷反应元件结合蛋白(CREB)、细胞外调节蛋白激酶(ERK1/2)蛋白磷酸化水平表达的影响。方法将25只SD大鼠,随机分为正常对照组、模型组、参归仁高、低剂量组(14.0、3.5 g·kg~(-1))、左洛复(盐酸舍曲林)对照组(4.5 mg·kg~(-1))。采用灌胃给药方式,连续30 d。分别取血、额叶组织,采用放射免疫法检测大鼠的E_2水平,采用免疫印迹法检测额叶p-CREB、p-ERK1/2的表达水平。结果与正常组比较,模型组大鼠血清中E_2、p-CREB、p-ERK1/2表达的水平明显降低;与模型组比较,参归仁高、低剂量组和左洛复组的额叶p-CREB的表达量明显升高,参归仁低剂量组和左洛复组的额叶p-ERK1/2表达量升高。结论参归仁合剂能够明显提高PPD大鼠的E_2水平,促进额叶p-CREB、p-ERK1/2的表达上调。
