目的研究酚红对无血清悬浮培养条件下肿瘤干细胞微球形成的影响。方法将人胶质瘤细胞U251、人乳腺癌细胞MCF-7、人肝癌细胞SMMC-7721、人宫颈癌细胞Hela等细胞分为酚红组和无酚红组,分别悬浮培养于添加了生长因子的DMEM-F12培养基中,观察5 d后肿瘤干细胞微球的形成情况,并计算微球的形成率。结果 U215无酚红组细胞的微球形成率为3.38%,明显高于酚红组的(2.73%);MCF-7、SMMC-7721、Hela无酚红组细胞的微球形成率分别为6.81%、7.75%、7.18%,酚红组的细胞均无微球形成。结论无血清悬浮培养肿瘤干细胞应采用无酚红培养基。
