白藜芦醇抗大鼠心肌缺血-再灌注后心律失常的机制研究

目的探讨白藜芦醇(Res)对大鼠心肌缺血-再灌注(I/R)后心律失常的影响及其与Ca2+-ATP酶(SERCA2)的关系。方法将SD大鼠随机分为假手术组、I/R后心律失常模型组、Res低、高剂量干预组(10、30 mg·kg-1,bid)。ig给予Res3 d后,制造I/R心律失常模型,用Power-Lab系统检测心电图的变化并进行心律失常评分,记录各组大鼠因心律失常导致的死亡数;用Real time PCR法检测心肌样本中肌浆网Ca2+-ATP酶中mRNA表达的变化。结果与模型组比较,Res组能显著减轻心室纤颤的发生率和持续时间,改善心律失常评分;上调SERCA2中mRNA的表达。结论 Res可明显减轻大鼠心肌I/R后心室纤颤,其机制至少可能与其减轻心肌细胞Ca2+超载相关。
