地西他滨对体外培养大鼠的血管平滑肌细胞ERα基因启动子甲基化状态及mRNA表达的影响

目的探讨地西他滨对大鼠主动脉平滑肌细胞雌激素受体α(ERα)基因甲基化水平及mRNA表达的影响。方法在体外培养的大鼠主动脉血管平滑肌细胞中加入0、1、2、4μmo.lL-1的地西他滨,孵育72 h,应用MS-PCR分析ERα基因启动子区域的甲基化状态,采用RT-PCR测定ERαmRNA的表达水平。结果用不同浓度的地西他滨处理大鼠血管平滑肌细胞后,其ERα基因启动子区域的甲基化水平降低,并在高浓度时出现去甲基化;ERαmRNA的表达增加并呈剂量依赖关系。结论地西他滨可促进大鼠血管平滑肌细胞ERα基因启动子区域去甲基化,并使ERαmRNA的表达增加。
