目的研究加替沙星(GFX)对人结肠癌HCT116细胞增殖和周期的影响,为研究GFX抗结肠癌的机制和作用靶点提供依据。方法采用CCK8法检测不同浓度(300~500μmol·L~(-1))GFX作用48 h对HCT116细胞增殖的影响;采用PI单染流式细胞技术检测不同浓度GFX作用48 h对HCT116周期的影响;Hoechst33258染色并用荧光显微镜观察GFX对HCT116形态的影响。结果与对照组比较,GFX各浓度组细胞的存活率下降,且呈剂量依赖性;400μmol·L~(-1 )GFX组的G_1期以及300、400μmol·L~(-1 )GFX组的S期细胞的比例均降低,300、400μmol·L~(-1 )GFX组的G_2期细胞的比例显著升高。荧光显微镜下未观察到经GFX处理的细胞存在明显的细胞凋亡特征。结论 GFX能抑制HCT116细胞的增殖并诱发其G_2期周期阻滞。
