目的 研究促凋亡蛋白BNIP3在涎腺腺样囊性癌(SACC)中的表达以及与临床病理特征及患者预后的关系,探讨其在腺样囊性癌低氧自噬中的作用。方法 采用免疫组织化学法检测65例SACC患者中BNIP3、低氧诱导因子(HIF)-1α和LC3的蛋白表达,分析BNIP3表达与SACC临床病理特征之间的关系及其与HIF-1α、LC3表达的相关性,并对患者生存率进行单因素生存分析。结果 BNIP3在41例SACC中阳性表达(63.1%),其表达的高低与组织学分级相关(P=0.001)。BNIP3与HIF-1α的表达相关(P=0.011),与LC3的表达无明显相关性(P=0.167)。BNIP3阴性表达组的总体存活率高于BNIP3阳性表达组(P结论 BNIP3在SACC的发生、发展中可能发挥着重要作用,为SACC的基因靶向治疗提供了新的治疗方向。

英文摘要

Objective This study investigated the expression of BNIP3 in salivary adenoid cystic carcinoma (SACC) and its correlations to the clinicopathological features and prognosis of patients with SACC. The role of BNIP3 in the progress of hypoxia-induced autophagy was elucidated.Methods The expression levels of BNIP3, hypoxia inducible factor (HIF)-1α, and LC3 in 65 SACC cases were detected by immunohistochemical staining method, and the correlation between the expression of BNIP3 and the clinicopathological features in SACC was analyzed. In addition, the correlations of BNIP3 gene expression with HIF-1α and LC3 gene expression were analyzed. The survival rate of patients with SACC was evaluated by univariate survival analysis.Results BNIP3 was considerably expressed in SACC in all three histological patterns, and was positive in 41 cases (63.1%). BNIP3 gene expression was significantly correlated with histological grade (P=0.001) and HIF-1α gene expression (P=0.011). By contrast, BNIP3 gene expression was not significantly correlated with LC3 gene expression (P=0.167). The overall survival rate of patients with negative BNIP3 expression was better than that of patients with positive BNIP3 expression (PConclusion BNIP3 might play a vital role in the tumorigenesis of SACC and may be a new target for gene therapy.
