利用毛细管电泳建立了中药苦蒿和青蒿及其制剂中11种酸性化合物的同时定量分析方法。其最佳电泳条件为:熔融石英毛细管50 cm(40 cm处检测窗口)×75 μm i.d.;分离电压:+15 kV;分离温度:25 ℃;缓冲液(含:35.7 mmol/L 磷酸钠,2.86×10-5mol/L 荧光素钠,少量甲醇,pH=11.42);进样压力:0.5 psi(3.45 kPa),进样时间5.0 s;荧光素钠为激光诱导荧光响应试剂;LIF激发光波长:488 nm,检测波长:520 nm。该方法线性范围宽、相对标准偏差低、精密度高、重现性好,用于实际样品的测定,获得满意结果。

英文摘要

A novel method was established to separate and quantify simultaneously 11 acidic compounds in traditional Chinese medicine of Conyza blinii levl. and Artemisia annua L. using capillary electrophoresis (CE). Optimized apparatus conditions were given. A fused-silica capillary 50 cm (40 cm to detector)×75 μm i.d. was used.Working voltage of +15 kV was applied at 25 ℃. The buffer(pH=11.42) containing 35.7 mmol phosphate sodium, 2.86×10-5mol/L fluorescein sodium (respondence reagent), and small amount of methanol was used. The hydrodynamic injection of a sample (0.5 psi(3.45 kPa)) during 5 sec was used. Indirect laser induced fluorescence(LIF) detection was applied by LIF detector at wavelength of 488 nm(excitation), 520 nm (detection).The method had a wide linear range, low relative standard deviation, high precision and perfect reproducibility. Real samples assay result was achieved perfectly in this condition.
