雷公藤红素与紫杉醇白蛋白纳米粒的制剂研究

目的制备雷公藤红素(CLT)与紫杉醇(PTX)共载的长循环白蛋白纳米粒,并对其进行表征和初步小鼠胰腺癌细胞的摄取研究。方法采用单因素筛选法依次筛选有机溶剂种类、白蛋白用量、DSPE-PEG用量、CLT与PTX的比例、大豆油和e80磷脂辅料的用量以及制备工艺,并对纳米粒粒径、电位、形态、包封率和载药量进行表征。同时应用流式细胞术对其细胞摄取情况进行研究。结果筛选出最优处方,成功制备了均一稳定的CLT与PTX共载长循环白蛋白纳米粒(CP-AN),其粒径约128 nm,PDI小于0.2,Zeta电位约-20 mV,对CLT的包封率约85%、PTX的包封率约75%。DSPE-PEG在一定程度上降低了细胞对纳米粒的摄取效率。结论 CLT与PTX具有良好的相容性,能应用简单的处方和超声工艺制成共载长循环白蛋白纳米粒。
